Typical use Of Kanamycin :Antimicrobial Spectrum Of Streptoycin Is Relatively Narrow, Active Primarily Against Aerobic Gram-negative Bacilli, But Potency Is Low. Sensitivie Organisms Are - H.ducreyi, Brucella, Yersinia Pestis, Francisella Tularensis, Nocardia, Calym, Granulomatis, M.tuberculosis, Only Few Strains Of E.coli, H.influenzae, V.cholerae, Shigella, Klebsiella, Entero-cocci And Some Gram-positive Cocci. Due To High Incidence Of Toxicity Both To Cochlea And Kidney It Is Now Occasionally Used Only As A Second Line Drug In Resistant Tuberculosis.
Mechanism of action of Kanamycin :Aminoglycosides Like Kanamycin "irreversibly" Bind To Specific 30S-subunit Proteins And 16S RRNA. Specifically Kanamycin Binds To Four Nucleotides Of 16S RRNA And A Single Amino Acid Of Protein S12. This Interferes With Decoding Site In The Vicinity Of Nucleotide 1400 In 16S RRNA Of 30S Subunit. This Region Interacts With The Wobble Base In The Anticodon Of TRNA. This Leads To Interference With The Initiation Complex, Misreading Of MRNA So Incorrect Amino Acids Are Inserted Into The Polypeptide Leading To Nonfunctional Or Toxic Peptides And The Breakup Of Polysomes Into Nonfunctional Monosomes.
Drug Interaction of Kanamycin :Kanamycin Is Known To Interact With Other Drugs Like Amphotericin B, Floxacillin (Na), Glucose, Pyridostigmine (Br). Always Consult Your Physician For The Change Of Dose Regimen Or An Alternative Drug Of Choice That May Strictly Be Required.
Side effects of Kanamycin :Nephrotoxicity, Ototoxicity, Eosinophilia, Vertigo, Headache, Nausea, Vomiting, Diarrhea, Tinnitus, Circumoral Paresthesia.
